The Tyneside club - a new addition to a reduced division next term - will host United on Wednesday, August 18, a week-and-a-half after the start of Leeds' Championship campaign.
East Division was a 10-club league last season but only eight sides will compete in it during the 2010-11 campaign, with Middlesbrough and Hull City the most high-profile opposition awaiting Leeds.
United's second string are scheduled to appear at Elland Road for the first time against Hartlepool United on Tuesday, August 24.
Under Academy manager Neil Thompson, Leeds have claimed the East Division title for two seasons running, retaining the prize they won in 2009 after denying Middlesbrough on the final day of last term.
